The amino acid pair, threonine and serine, are separated by ion exchange column chromatography, e.g. on a polystyrene cation exchanger, using an eluent mixture, which contains more than I %, preferably 3-8%, by volume of a glycol or a glycol alkyl ether, such additive having a boiling point above 100‹C. The additive is suitably ethylene glycol monomethyl ether. Where a series of amino acids is to be separated, the additive-containing eluent is preferably employed from the time of introduction of the sample to be separated to the column up to the final separation of serine; thereafter eluent without the additive is employed. About 5% thiodiglycol may be included in the eluent as antioxidant. The separation of the tyrosine-phenylalamine, glycine alamine, and isoleucine-leucine pairs is also stated to be improved.
