1280779 1-Oxa-4,8-diazaspiro[4,5]decanes; 1 - oxa - 5,9 - diazaspiro[5,5]undecanes YOSHITOMI PHARMACEUTICAL INDUSTRIES Ltd 18 Dec 1970 60322/70 Heading C2C The invention comprises compounds of formula and their pharmaceutically acceptable acid addition salts, wherein R is H, acetyl, allyl, propargyl, 2-cyanoethyl, C 1-4 alkyl, C 1-4 alkoxycarbonyl, C 1-4 alkoxycarbonylethyl, PhCO, PhCH 2 or PhCH 2 CH 2 ; RSP1/SP, RSP2/SP are each H, Me, Et or Ph; RSP3/SP is H, C 1-4 alkyl, PhCH 2 , or Ph (optionally substituted by C1, Me, MeO or CF 3 ); and n is 0 or 1. These compounds may be prepared by (1) reaction of a 1-R-4-piperidone with or with and RSP3/SPNH 2 ; (2) introduction of the R group (other than H) with the above compound where R is H; (3) forming the compound where R is H by hydrogenolysis of the compound where R is PhCH 2 , or by hydrolysis of the compound where R is C 1-4 alkoxycarbonyl. Therapeutic compositions for oral or parenteral administration comprise compounds of the above formula, which are reserpine antagonists and blood sugar lowering agents.
