In determination of a ligand or the ligand-binding capacity in a liq. medium by specific binding in which the liq medium is brought into contact with a determination reagent comprising labelled binding component to form a reaction system in which the bound and the free from of the labelled component are present, the bound form is sepd. from the free form of the labelled component, and the labelling of the bound or free form of the labelled component is determined. The sepn. is carried out by selective absorption of the free form of the labelled component on a solid comprising a non-ion exchanging crosslinked PVA. The procedure is partic. suitable for the radioimmunological determination of non-protein haptens such as thyroxine, and triiodothyronine, oestriol, vitamin B12 and digoxin in biological fluids such as serum. The crosslinked PVA is a very effective and readily obtainable material for carrying out the sepn. of bound and unbound ligand.
