PRODUCTION OF PHOTOPROTEIN AEQUORIN USING EXTRACELLULAR SECRETORY VECTOR OF ESCHERICHIA COLI

PURPOSE:To make it possible to accumulate aequorin protein outside a cell, by cloning cDNA clone plasmid of photoprotein aequorin into an extracellular secretory vector to give a plasmid and integrating the plasmid into Escherichia coli. CONSTITUTION:cDNA clone plasmid pAQ440 of photoprotein aequorin is connected to a gene for secretion of adventitial protein of Escherichia coli and promoter of lipoprotein (lpp) to construct a secretory manifestation vector to release aequorin protein to the outside of cell of Escherichia coli. By using the vector, aequorin protein produced in the cell of Escherichia coli is released to the outside of the cell of Escherichia coli.
