PURPOSE:To analyze monensin with a high accuracy, by separating a sample obtained in a specified process with a reversed phase distribution column with methanol as mobile phase to detect fluorescence. CONSTITUTION:An extraction liquid obtained from a sample is solidified by drying. A solid is dissolved by a 0.02-0.1mol. hydrogen chloride solution and chloroform equivalent thereto. A chloroform solution of n-hexadecanic acid is added to the solution by a known amount as internal reference substance and after a fixed time of leaving the solution alone, a chloroform layer is sampled to dehydrate. 3-5 fold mol. of a chloroform solution of 9-anthryl diazomethane or 9-butyl 10-hydroxymethylanthracene is added to the solution by an equal amount. The sample obtained is prepared to detect fluorescence using a reversed phase distribution column with methanol or a mixed liquid methanol and tetrahydrofuran as mobile phase thereby achieving highly accurate analysis of monensin.
