PURPOSE:To obtain a novel DNA at a replication initiating point of an animal DNA by subjecting an Escherichia coli plasmid to specific treatment with a mammalian DNA and restriction enzyme DpuI, transforming Escherichia coli and collecting the objective DNA. CONSTITUTION:A mammalian DNA is inserted into an Escherichia coli plasmid and the resultant many recombinant plasmids are propagated in Escherichia coli and the propagated plasmid DNA is then Propagated in an animal cell. The above-mentioned DNA is subsequently propagated in an animal cell and then treated with a restriction enzyme DpuI to transform Escherichia coli and collect the DNA. Thereby, the objective DNA is obtained. A restriction enzyme cleavage map of the DNA obtained by the afore-mentioned method is expressed by figure I. The part of the replication initiating point in the DNA is considered as present in the 633rd to the 666th residues. The base sequence thereof is indicated by the formula.
