PURPOSE:To obtain the titled plasmid useful as a shuttle vector in case a coliform bacillus or blue-green alga is used as a host, by bacteriolyzing Anacystis nidulans by the enzymic method, and separating and purifying the bacteriolyzed Anacystis nidulans by the density gradient centrifugation method. CONSTITUTION:Plasmid pBA1 obtained by bacteriolyzing a blue-green alga Anacystis nidulans having about 5 megadaltons molecular weight and a map for cleavage by restriction enzymes shown in the figure by the enzymic method, and separating and purifying the resultant plasmid by the density gradient centrifugation method. The specific positions of the plasmid are cleaved by a restriction enzyme and a desired exogenote DNA is integrated and joined with a ligase to regenerate a cyclic plasmid, which is then inserted in a coliform bacillus or blue-green alga to expect the multiplication in a new host-vector system or development of a gene.
