CONSTRUCTION OF EXPRESSION PLASMID PAMC1 FOR EXPRESSION OF RAT HEPATIC CYTOCHROME P-450MC GENE IN YEAST

摘要

PURPOSE:To enable the expression of cytochrome P-450MC having high hydroxylation activity and wide substrate specificity in a yeast, and to apply the plasmid to an oxidation reaction and to the oxidative removal of organic compounds in waste water. CONSTITUTION:The cytochrome P-450MC gene separated from the recombinant plasmid PAU157 having whole coding region of the rate hepatic cytochrome P-450MC protein, is integrated into the expression vector PAAH5 holding an alcohol dehydrogenase promoter. The expression plasmid PAMCI constructed by the above process is introduced in a host yeast, and the expression of the gene is carried out in large amount. A large amount of pure rat hepatic cytochrome P-450MC enzyme can be separated and purified by this process.
