PURPOSE:To enable the accurate determination of a decomposition enzyme of a peptide having leucine at the N-terminal, by using O-phenanthroline as an antienzyme, thereby separating serum leucine aminopeptidase from the other decomposition enzyme. CONSTITUTION:O-phenanthroline is used as an antienzyme for the quantitative determination of serum leucine aminopeptidase using a substrate consisting of a peptide having leucine at the N-terminal. The enzymatic activity of allyl amidase and cystine aminopeptidase which are hydrolase of the same peptide can be inhibited and, accordingly, the serum leucine aminopeptidase can be exclusively determined by selecting the amount of O-phenanthroline added to the system. The determination is preferably carried out at an O-phenanthroline concentration of 0.9-5.5mM and 8.0-9.5pH.
