PURPOSE:To efficiently produce a thermostable lipase by introducing a gene capable of coding the thermostable lipase into a vector DNA, transforming a host cell with the resultant recombinant vector and culturing the obtained transformant. CONSTITUTION:A chromosomic DNA, containing a thermostable lipase gene and obtained from a microorganism Pseudomonas KWI-56 strain (FERM P-9659) capable of producing the thermostable lipase is isolated. The resultant chromosomic DNA and vector DNA are cleaved with a restriction enzyme and both are then mixed and treated with a DNA ligase to prepare a recombinant vector, which is used to transform Escherichia coli, Bacillus subtilis, etc. the obtained transformant is subsequently cultured under the optimum conditions for producing the enzyme. The thermostable lipase is then recovered from the resultant culture supernatant or crushed microbial cells.
