TOMATO ACIDIC INVERTASE COMPLEMENTARY GENE, RECOMBINANT VECTOR HAVING THE SAME GENE AND TRANSFORMER HAVING THE SAME RECOBMINANT VECTOR

PURPOSE: To obtain a new cDNA useful for increasing or suppressing acidic invertase activity in microorganisms, plants and animals. ;CONSTITUTION: A cDNA coding tomato acidic invertase of a base sequence of the formula. The cDNA prepared from tomato fruit mRNA is bound through an adaptor DNA having EcoRI part to EcoRI part of λgt10 phage vector with a ligase and then, phage particles are formed to prepare a cDNA library. Screening of acidic invertase gene from this library is carried out with a probe obtained by labelling an oligonucleotide synthesized based on decision of partial amino acid sequence of acidic invertase with ³²P and phage DNA IS extracted from phage particles obtained by large-scale culture of the resultant transformer and further digestion of EcoRI and electrophoresis of agarose gel are carried out to provide a cDNA fragment corresponding to the acidic invertase.;COPYRIGHT: (C)1994,JPO&Japio
