摘要

PURPOSE: To obtain a new cDNA useful for increasing or suppressing acidic invertase activity in microorganisms, plants and animals. ;CONSTITUTION: A cDNA coding tomato acidic invertase of a base sequence of the formula. The cDNA prepared from tomato fruit mRNA is bound through an adaptor DNA having EcoRI part to EcoRI part of λgt10 phage vector with a ligase and then, phage particles are formed to prepare a cDNA library. Screening of acidic invertase gene from this library is carried out with a probe obtained by labelling an oligonucleotide synthesized based on decision of partial amino acid sequence of acidic invertase with 32P and phage DNA IS extracted from phage particles obtained by large-scale culture of the resultant transformer and further digestion of EcoRI and electrophoresis of agarose gel are carried out to provide a cDNA fragment corresponding to the acidic invertase.;COPYRIGHT: (C)1994,JPO&Japio
