Chinese hamster ovary (CHO) cells have been widely used as a host for the production of therapeutic monoclonal antibodies. Rretrotransposon can transfer to new genomic loci by a “copy-and-paste” process known as retrotransposition (Fig. 1). A full-length long interspersed element-1 (LINE-1, L1) contains a 5’ untranslated region (5’UTR), two non-overlapping open reading frames (ORFs) that are separated by a short inter-ORF sequence, and a 3’UTR that ends in an adenosine-rich tract. Previously, we focused on the ability of retrotransposition to transfer transgene into the CHO genome and we also reported the establishment of recombinant CHO cells using LINE-1 as a vector 1 . Here, we evaluated the characteristics of recombinant CHO cells producing an anti-prion single chain Fv fused with Fc region (scFv-Fc).
