Process for preparation of a protein mitogenica B induced by the African swine fever virus

Mitogenic B protein induced by the virus of African swine plague, having isoelectric pt. 3.5-4.3, optical absorption max. at 270-290 microns stainable by Coomassie blue, mol. wt. 25000-35000 clattons and denaturable by heating to around 70 deg.C for around 10 mins. Its prepn. consists of (a) obtaining porcine leucocytes in supernatant plasma of sedimented erythrocytes; (b) mixing this with a suitable culture medium, buffered and supplied with antibiotics; (c) culture of the mixt. followed by removal of non-adherant cells; (d) infection of teh monocytes from (c) with Frican swine plague virus and culture in a suitable buffered medium supplied with antibiotics; (e) removal of the contaminant virus by centrifuging and concn.; and (f) successive fractionation and purificn. to obtain an active and pure protein prod.
