A method (A) for determn. of 1,5-anhydroglucitol (1,5-AG) in a sample comprises: (a) treating the sample with an alpha-glucosidase (I) derived from a Bacillus sp. to convert maltose to glucose; (b) selectively removing the glucose and other sugars (other than 1,5-AG) from the sample; and (c) quantitatively determining the 1,5-AG concn. using a pyranose oxidase (PROD). Also claimed is a method (B) for determn. of 1,5-AG in a sample, comprising: (a) treating the sample with (I) to convert maltose to glucose; (b) phosphorylating the glucose with hexokinase and excess ATP at pH 7.2-8.5, whereby (sic) the glucose is selectively removed together with other sugars (other than 1,5-AG); and (c) quantitatively determining the 1,5-AG concn. by direct reaction with a PROD at pH 7.2-85. USE/ADVANTAGE - The methods may be used for diagnosis of diabets. Errors associated with the presence of maltose in the sample are eliminated. The methods are simple, quick and suitable for automatic analysers.
