EGG YOLK TREATMENT METHOD FOR MEASURING AVIAN INFECTIONS BURSA FABRICII DISEASE VIRUS ANTIBOSY IN EGG YOLK

PURPOSE:To measure an avian infectious bursa fabricii virus antibody (IBDV) by simple operation by putting a dilute soln. prepared by diluting egg yolk of a chicken with a dilution soln. by 2-3 times in a container and shaking the container until obtaining uniform concn. to prepare a specimen for measuring an antibody according to a latex agglutination method. CONSTITUTION:A mouse is immunized with purified IBDV to form a monoclone antibody and antibody sensitized latex particlses for detecting IBDV are prepared. For example, a hybridoma producing this antibody is injected into the abdominal cavity of a nude mouse to sample abdominal dropsy containing the antibody in high concn. This abdominal dropsy is mixed with the latex particles under stirring to be reacted therewith. After reaction, the particles are settled to be suspended in a phosphate buffer soln. to obtain antibody sensitized latex particles for detecting IBDV. Next, bursa of Fabricius is extracted from a chicken infected with IBDV several days after infection and an equal amt. of a phosphate soln. is added to the bursa of Fabricius and the resulting mixture is homogenized to obtain an IBDV standardized antigen which is, in turn, mixed with egg yolk and the particles to measure an antibody on the basis of the obstruction of agglutination.
