PROBLEM TO BE SOLVED: To regenerate all the varieties including the elite varieties of a low regeneration rate and to convert their characters as the result by utilizing immatured cotyledon cell at the time of regenerating Medicago Sativa, so as to improve its regeneration rate. SOLUTION: At the time of regenerating Medicago Sativa, the somatic embroyogenesis of the immatured cotyledon cell of Medicago Sativa is started at first. The immatured cotyledon cell is desirably a half-transparent to light green immatured cotyledon cell ten days after pollination. In addition concerning the somatic embroyogenesis, it is desirable that after the extorpation of the immatured cotyledon from the seed embryo of Medicago Sativa, the cotyledon is brought into contact with auxin to guide the cell division and the cell growth of the cotyledon. In addition, at the time of regenerating the variety of the Medicago Sativa which can not be regenerated in a conventional method, it is desirable to culture the somatic embryo in a plant which can be regenerated after starting the somatic embroyogenesis of the immatured cotyledon cell.
