The present invention relates to an improved reagent composition and method for accurately and precisely differentiating leukocytes and analyzing subgroups using fresh or aged blood samples. The present invention is particularly applicable to the analysis of aged blood samples that have been stored at room temperature for more than one day, thus providing accurate and useful information from samples that are considered to be less than optimal. The improved reagent compositions and methods relate specifically to peroxidase and methods for distinguishing leukocytes. One aspect of the invention includes improved aqueous reagent compositions for performing peroxidase methods to differentiate. Other aspects include the use of a rinsing solution and rinse cycle without hemolytic surfactant to mitigate the adverse effects of rinsing residues and to rationalize and economize the analysis process, particularly when the analysis is performed on automated hematology analyzers and flow cytometry systems do. The compositions and methods of the present invention provide clinically useful data for differential analysis of whole blood samples.
