-34-The present invention relates to an in vitromammalian DNA replication system and to a method foridentifying agonists and antagonists of DNAreplication. The method comprises contacting in vitroa plasmid with a mixture comprising nuclear orcytoplasmic extracts from HeLa cells, the drug and amixture of nucleotides, assessing the stimulation orthe inhibition of initiation of DNA replication andthe elongation of nascent DNA produced by the drug,and identifying the essential structures of the drugby quantitative structure-activity relationship (QSAR)analysis deriving relationships between the structuralfeatures of the drug and biological responses producedby the binding of the drug to the target receptor.The plasmid has a target receptor and comprises aspecific mammalian origin of DNA replication.
