PROBLEM TO BE SOLVED: To provide a method for observing ligand-specific opening of a gated- porin channel in an outer membrane of a living bacterium usable for therapeutic drug delivery or the like by a spin-labeling membrane transport site, and measuring the conformational change of the membrane transport site. ;SOLUTION: A membrane transport site is spin-labeled to provide a spin- labeled membrane transport site, and the conformational change of the spin- labeled membrane site is measured to observe the in-vivo membrane transport in a bacterium. Preferably, the residue of the bacterium on the surface of an iron channel is modified to provide the modified residue of the bacterium, and the modified residue of the bacterium is labeled to provide the labeled and modified residue of the bacterium. The conformation changes in the labeled and modified residue of the bacterium and the iron-bound protein are measured while the labeled and modified bacterium is reacted with an iron-bound protein to observe the in-vivo molecular transport during the iron channel transportation of the living bacterium. A therapeutic agent can be sent into a human body by the iron transport activity in the bacterium.;COPYRIGHT: (C)1999,JPO
