PURPOSE: Provided are a novel microorganism, Penicillium simplicissimumGM31, isolated from soil and producing beta-D-glucosaminidase used for the manufacture of D-glucosamine and a process for preparing D-glucosamine by using the same protein. CONSTITUTION: The manufacturing method of beta-D-glucosaminidase having 110000 Da of molecular weight comprises the steps of: culturing Penicillium simplicissimumGM31 (KMCC-10215) in MCY medium containing 1.0-5.0%(w/v) of colloidal chitosan at 20-30 de3g.C and pH 5.0-9.0; and subjecting the culture to column chromatography. D-glucosamine is effectively manufactured by decomposing chitosan with the manufactured protein, beta-D-glucosaminidase.
