ISOLATION AND PURIFICATION METHOD OF SAPONIN MUTANT QS-L1 FROM QUILLAJA SAPONARIA MOLINA

PURPOSE: An isolation and purification method of saponin mutant QS-L1 from Quillaja Saponaria Molina is provided to easily and effectively purify a large amount of QS-L1 saponin fraction, so that it can be useful as an immune adjuvant. CONSTITUTION: The isolation and purification method of saponin mutant QS-L1 from Quillaja Saponaria Molina comprises the steps of: dissolving the surface extract of Quillaja Saponaria Molina in distilled water and centrifuging the solution to remove insoluble substance and collect the supernatant; subjecting the supernatant to C4 reverse-phase high pressure liquid chromatography(HPLC) to collect an active fraction; and subjecting the active fraction to silica gel column chromatography, wherein the C4 reverse-phase HPLC uses an eluting agent having acetonitrile density gradient; and the silica gel column chromatography uses an eluting agent wherein chloroform, methanol and distilled water are mixed in a ratio of 62 to 32 : 32 : 6 (v/v).
