The procedure is for the production of a polypeptide in yeast and involves a first stage of fermentation of a yeast cell containing a sequence of deoxyribonucleic acid codifying a polypeptide of interest, which is transcribed under control of a promoter of pH under conditions of pH permitting cellular growth. In a second stage, the pH value is increased to augment the transcription of the polypeptide. In a third stage, fermentation occurs during a determined period of time and under the same conditions of pH as the previous stage. In a fourth stage, the polypeptide produced is isolated.
