A novel expression cloning method for detection and identification of proteinscapable of binding to tyrosine-phosphory-lated domains of receptor tyrosine kinases is based on the use of a novelprobe. This probe comprises an amino acid sequence de-rived from the tyrosine-phosphorylated-portion of the receptor molecule, or afunctional derivative thereof, and has at least onephosphorylated tyrosine residue, lacks the tyrosine kinase portion of thereceptor, and is delectably labeled. Also disclosed are amethod for preparing the probe, a method for mapping to a chromosome a geneencoding a protein capable of binding to tyro-sine-phosphorylated domains of receptor tyrosine kinases, and a method forpurifying such a protein with the probe. Two pro-teins discovered using the above cloning method, GRB-1 and GRB-2, DNA encodingthese proteins, and methods for detectingthese proteins are also disclosed.
