It is found out that a template DNA sample from which impurities other than DNA have been removed to such an extent as causing no problem in a reaction can be prepared by adhering test microbial cells to a water-absorbing support, then growing the cells, treating the support with a mixture of equal parts of phenol and CIA, centrifuging and collecting a definite amount of the supernatant followed by evaporation to dryness. According to this method, it is also possible to replicate and store a template DNA sample prepared under the same conditions. When the template DNA prepared by this method is employed in PCR, gene amplification products can be efficiently obtained in a large number of fungi and yeasts.
