FIELD: biotechnology, molecular biology, genetic and protein engineering, microbiology.;SUBSTANCE: invention relates to a method for transformation designated for expression of heterologous proteins in yeast Candida utilis. Method involves preparing auxotrophic mutants of indicated species and isolation of genome library of different genes complementing indicated auxotrophic mutants. As host-cells the indicated transformation system uses new auxotrophic strains obtained from yeast of the strain Candida utilis NRRL Y-1084 (with phenotype ura and phenotype his denoted as TMN3) wherein biosynthetic pathways in metabolism of uracil and histidine are absent and these strains can be transformed with plasmids comprising genes Candida utilis URA3 and HIS3 as selective markers. Other objects of invention relate to recombinant DNA material for transformation of indicated strains and gene HIS3 DNA encoding sequence in indicated yeast strains.
