Described is method for testing samples containing prion protein for the presence of the PrPSc form. The method comprises adding a protease to a sample to digest protease-sensitive proteins or regions of protein, then after digestion testing for the presence of the prion protein region PrP 27-30, which is protease-resistant in the PrPSc for the prion protein, and testing the sample for whether the protease-sensitive region of the prion protein is digested. The detection of PrP 27-30 indicates the presence of PrPSc in the sample.
