PURPOSE: Provided are composition for detecting β-1,3-glucan existing in an infinitesimal quantity in a sample, a method for preparing the same and a kit article for detecting β-1,3-glucan. CONSTITUTION: A composition for detecting β-1,3-glucan is prepared by the following steps of: (a) obtaining a mixture of a body fluid or plasma and globule hemolytic material from insects in the presence of a chelating agent; (b) obtaining a fraction by treating them with solvent or buffer solution including sufficient chelating agent for chelating calcium ion left in separation process; and (c) selecting the fraction having phenoloxydase activity by β-1,3-glucan in the presence of calcium.
