The present invention provides a hemoglobin (Hb) molecule having six sym one PEG chains, where two of said PEG chains are linked to Hb Cys-93 (225), and the remaining PEG chains are linked. to thiol groups introduced into Hb sym -NH-2 ~. The present invention also provides a process for the preparation of a modified hemoglobin (Hb) molecule comprising the steps of: (a) reacting HB with an 8-15 fold excess of iminothiolane to form thiolated Hb; and (b) reacting thiolated Hb with a 16-30 fold excess of functionalized PEG with a maleimide moiety to form the modified Hb.
