摘要

It is intended to provide a novel enzyme which is a recombinase irreversibly inducing a site-specific gene recombination and an irreversible site-specific gene recombination method using this enzyme. The above method can be carried out as follows: combining (i) a polynucleotide having a recognition sequence (attP1/P2) wherein a first recombination site (attP1) and a second recombination site (attP2) are connected via a core sequence, (ii) a polynucleotide having a recognition sequence (attB1/B2) wherein a third recombination site (attB1) and a fourth recombination site (attB2) are connected via a core sequence and (iii) an integrase originating in a Staphylococcus aureus bacteriophage ΦMR11 to thereby form (iv) a polynucleotide having a recombined sequence (attP1/B2) wherein the first recombination site (attP1) and the fourth recombination site (attB2) are connected via a core sequence and (v) a polynucleotide having a recombined sequence (attB1/P2) wherein the third recombination site (attB1) and the second recombination site (attP2) are connected via a core sequence.
