An RNA-cleaving DNAzyme is provided to cleave a target RNA substrate and be used for identifying a DNAzyme target site in an RNA molecule by using DNAzyme library containing the DNAzyme. An RNA-cleaving DNAzyme includes a catalytic core of a sequence described as SEQ ID : NO. 1; a substrate-linked arms having a random nucleotide sequence adjacent to the catalytic core; and a fixed 5'/3' flanking sequence for amplification, wherein the RNA is a gene coded by a virus gene and the DNAzyme is described as SEQ ID : NO. 2, 3, 4, or 5.
