METHOD FOR QUANTITATIVE DETECTION OF HUMAN FETAL HEMOGLOBIN

FIELD: medicine, laboratory diagnostics.;SUBSTANCE: quantitative detection of human fetal hemoglobin (HbF) should be carried out immunologically in gel carrier with specific antibodies to fetal hemoglobin. The method suggested should be fulfilled with the help of rocket electrophoresis in agar gel at the ratio of 20 ml gel/0.5 ml antiserum; the samples under testing should be pre-mixed with 0.2%-aqueous solution of sodium dodecyl sulfate (SDS) at equal volumetric proportion, dried plates of gel obtained as a result of electrophoresis should be stained with 0.3%-guaiacol solution upon 0.1 M acetate buffer at pH=4.6 with 0.2% MnCl₂ and 0.1% hydrogen peroxide. One should detect the concentration of HbF according to standard calibrating curves. Application of the present method enables to increase the accuracy of quantitative HbF-detection.;EFFECT: higher efficiency.;2 dwg, 5 ex
