A method of manufacturing a hydroxy linoleic acid is provided to control various distributions of the hydroperoxy linoleic acids using a lipoxygenase enzyme originated from Indian corn. A method of manufacturing a hydroxy linoleic acid comprises steps of: performing the double location specific hydroperoxidation reaction about a linoleic acid using a mixing solution including the linoleic acid and a lipoxygenase enzyme originated from Indian corn(Zea mays lipoxygenase1: LOX1) which is a protein of a sequence number 2 or a protein having a sequence homology of 70%% or more with the protein of the sequence number 2; reducing a reaction product which is 13-(9Z,11E)-hydroperoxy octadecadienoic acid, 13-(9E,11E)-hydroperoxyoctadecadienoic acid, 9-(10E,12Z)-hydroperoxy octadecadienoic acid or 9-(10E,12E)-hydroperoxy octadecadienoic acid by using a triphenylphosphine(TPP) which is a reducing agent or an NaBH4; separating a reduced matter which is 13-(9Z,11E)-hydroxy octadecadienoic acid, 13-(9E,11E)-hydroxy octadecadienoic acid, 9-(10E,12Z)-hydroxy octadecadienoic acid or 9-(10E,12E)-hydroxy octadecadienoic acid. A concentration of the linoleic acid is 0.03mM~0.5mM. A concentration of the lipoxygenase enzyme is 0.4 mum/ml~28.8 mum /ml.
