In order to examine an object comprising fluorophores, and having a first face and an opposite second face, the method comprises a first step of illumination (f1) of the first face of the article with an excitation light of the fluorophore and a first step of acquisition of a first (f2) image by the detection of a light emitted by the second face of the object. In addition, the method comprises a second step of illumination (f3) of the second face of the article with an excitation light of the fluorophore and a second step of acquisition of a second image (f4) by détectior of a light emitted by the first face of the object. The reconstruction of the distribution of fluorophores in the object (f5) is carried out by means of the first and second image.
