PURPOSE: A knock-out vector which is prepared using a yellow fluorescent protein reporter knock-in vector is provided to stably and effectively express a reporter by binding ORF with a promoter sequence of knock-out gene.;CONSTITUTION: A yellow fluorescent protein (YFP) reporter knock-in vector comprises a sequence of the sequence number 1. A method for manufacturing the knock-out vector containing the YFP reporter gene comprises: a step of mutating the YFP gene sequence containing ATG as an initiation codon to prepare the ORF of YFP gene; a step of preparing a promoter sequence of the gene and polynucleotide to knock out; and a step of binding the promoter sequence and polynucleotide to the 5' terminal of ORF sequence of YFP.;COPYRIGHT KIPO 2010
