METHOD FOR EXTRACTING NUCLEIC ACID OF RALSTRONIA SOLANACEARUM BY REMOVING A MATERIAL WHICH REMOVES NUCLEIC ACID POLYMERIZATION

PURPOSE: A method for extracting nucleic acid of Ralstonia solanacearum which is a plant pathogenic microorganism is provided to improve extraction efficiency by Guanidine thiocyanate method.;CONSTITUTION: A method for extracting and purifying nucleic acid from soil microorganism sample comprises: a step of centrifuging 1ml of soil microorganism culture medium to obtain cell pellet; a step of washing three times with phosphate buffer saline(PBS) and resuspending in 200ul of PBS; a step of adding 700ul of Guanidine thiocyanate to 300ul of washed sample and mixing for 10 minutes; a step of injecting 500ul of isopropyl alcohol and centrifuging to obtain DNA pellet; a step of adding 200ul of chelex-100 resin to DNA pellet and boiling 10 minutes; and a step of centrifuging.;COPYRIGHT KIPO 2010
