A transformation system for introduction of Podophyllum peltatum gene using Agrobacterium tumefaciens is provided to produce a large quantity of podophyllotoxin as an anticancer agent by introducing a gene encoding podophyllotoxin into the calluses of Podophyllum peltatum by coculturing with Agrobacterium and subculturing a small amount of transformed calluses. The transformation system for introduction of Podophyllum peltatum gene using Agrobacterium tumefaciens comprises the steps of: inducing somatic embryo-generating calluses of Podophyllum peltatum from cotyledon of Podophyllum peltatum in a callus-inducing medium; transforming Agrobacterium GV3101 with a vector pBI 121 containing the gene encoding podophyllotoxin; coculturing the transformed Agrobacterium GV3101 with the embryo-generating calluses of Podophyllum peltatum and selecting the transformed embryo-generating calluses; and regenerating the transformed embryo-generating calluses.
