DNA extension and analysis using a rolling primer

PROBLEM TO BE SOLVED: To provide a method for determining a base sequence by providing a set of primers, forming a template containing a primer binding site, etc., amplifying and identifying a double stranded DNA in the extension of the primer, and shifting the primer binding site, etc. ;SOLUTION: This method for determining a base sequence comprises providing a set of primers each having an extension region containing a terminal nucleotide, a segment arranged on a template and a nucleotide having one or more less complexity, forming a template containing a primer binding site which is complementary to at least one of the primers of the above set and a polynucleotide, amplifying a double stranded DNA by extending the primer in the extending region for forming the double strand completely matching with the primer binding site of the template, identifying the nucleotide at the terminal of the primer by identifying the amplicon, shifting the binding site by one base by mutating the primer binding site of the template, and repeating these processes for determining the sequence of the polynucleotide.;COPYRIGHT: (C)1999,JPO
