The invention relates to a process for obtaining lipase with a Yarrowia lipolytica strain by submerged cultivation of the microorganism on a culture medium with mineral salts, glucose, peptone, yeast extract and sunflower oil or olive oil, followed by extraction and purification of the enzyme, where the sunflower oil or the olive oil is contained in the fermentation medium in a concentration ranging from 0.05 to 0.1%.
