The present invention provides a method of site-directed mutation of nucleic acid sequence, which includes designing forward primer in the left side of the center point of the sequence, designing backward primer in the right side of the center point of the sequence, and designing a center primer comprising the center point of the sequence; and then mixing forward primer, backward primer and the center primer, and carrying out overlapping PCR. The method can be used for screening monoclonal antibodies.
