Method for determining methylation of a nucleic acid comprising the following steps: a) fragmentation of a genomic DNA sample with at least one enzyme methylation sensitive restriction and at least one restriction enzyme methylation insensitive, b) ligation, at the ends of the DNA fragments obtained, of specific adapters where one of the specific adapters comprises a promoter sequence functional, c) amplification of the fragments that include both adapters using specific primers based on the adapters, d) labeling the fragments amplified by in vitro transcription with an RNA polymerase capable of initiating transcription from the promoter sequence contained in one of the adapters using a mixture of DNA nucleotides and e) determining the methylation status of the sample.
