The present invention relates to a method for the simultaneous detection and identification in a sample of at least one species of the family Engraulidae selected from Engraulis encrasicolus, Engraulis mordax and Engraulis ringens, by PCR amplification multiplex of at least one of sequences, selected from SEQ ID nO 1, SEQ ID nO 2 and SEQ ID nO 3, corresponding with the spacer not transcribed (NTS) DNA 5S ribosomal at least one of the three species, using at least one of the pairs primers sequence selected from: SEQ ID NO 4 and SEQ ID NO 5; SEQ ID NO 6 and SEQ ID NO 7; and SEQ ID NO 8 and SEQ ID NO 9.
