PURPOSE: A microorganism with improved productivity of FK506 or FK520 and a mass-production method of FK506 or FK520 using the same are provided to be applied in mass production strains of FK 506 or FK 520 by investigating Tcs7 genes and FkbN genes which are positive regulators. CONSTITUTION: For FK506(tacrolimus) or FK520(ascomycin) producing microorganisms, activation of Tcs7 protein becomes weaker than inherent activation of wild-type microorganisms in KCTC 11604BP which belong to Streptomyces sp. The protein activation of the microorganisms becomes weaker by deletion or mutation of the Tcs7 genes which code the Tcs proteins. The Tcs7 protein has an amino acid sequence of sequence number 1, and the Tcs7 gene has a base sequence of sequence number 2. Additionally, activation of FkbN proteins is strengthened compare to the inherent activation of the wild-type microorganisms.
