Invention relates to apple and pear propagation using in vitro cultures of micro-propagation by methods of biotechnology. The invention offers a 2-step method for assessment of somaclonal variation, based on microsatellite (SSR) and methylation-sensitive amplified polymorphism (MSAP) molecular markers. The initial assessment of somaclonal variation is applicable for all stages of in vitro propagation, whereas detailed assessment is applicable in the final phase of in vitro propagation to ensure the identity of reproduced material with the initial plant material.
