The present invention relates to a process of preparing a silk fibroin membrane for culturing human cells, the silk fibroin membrane prepared by the process, and a composition containing the silk fibroin membrane for culturing human cells and, more particularly, to a process of preparing a silk fibroin membrane by adding a crosslinking agent to silk fibroin proteins extracted from cocoons and irradiating with ultrasonic waves. The silk fibroin membrane prepared by the process has strong fiber bond and low dissociation possibility during cell culture, thereby effectively culturing various cells due to the curve on the surface.
