PROBLEM TO BE SOLVED: To provide a nucleic acid amplification method and a method for producing a nucleic acid sample for nucleic acid amplification which can collect nucleic acids simply and efficiently in a short time, without requiring enzyme treatment for tissue lysis in extracting nucleic acids from a biological tissue.SOLUTION: The method of the invention is characterized by comprising a heat treatment step of heating a mixture of an isolated biological tissue and an aqueous solvent, and releasing nucleic acids from the biological tissue; and an amplification step of performing nucleic acid amplification in the reaction solution comprising a nucleic acid sample, using the mixture after the heat treatment step as the nucleic acid sample, and using nucleic acids in the nucleic acid sample as a template. In the heat treatment step, the heat treatment time is 5 to 60 minutes. In the amplification step, the nucleic acid sample comprised in the reaction solution per 50 μL is obtained from 0.01-1 mmof the total volume of the biological tissue. In before and after the heat treatment step, enzyme treatment by an enzyme for tissue lysis is not performed.
