An amplification method based on gene saltatory replication by nucleic acid isothermal amplification. A designed primer, under the effect of Bst DNA polymerase, can complete high specificity, effective, and rapid DNA amplification under isothermal conditions. After the primer fuses with a target DNA, the primer does not completely undergo mutual extension amplification according to the target DNA sequence, but undergoes saltatory replication during the amplification process, such that the amplified product segment is different from the anticipated product segment, in other words, differing from the amplification products of other nucleic acid amplification methods.
