Companion diagnostics for TEC family kinase inhibitor therapy

The present invention provides methods, assays, and systems for determining the effectiveness of TEC family kinase inhibitors against target kinases. The methods, assays, and systems relate to determining the occupancy of a target kinase by a TEC family kinase inhibitor (eg, a BTK inhibitor). Such quantitative measures are used to provide information about the subject's treatment and overall health care. For example, a diagnostic kit is provided for diagnosing, predicting, and monitoring a disease, or for demonstrating that an effect of treatment with a TEC family kinase inhibitor is expected. In another example, diagnostic kits are also provided for identifying responses to TEC family kinase inhibitor therapy, determining treatment regimens, and detecting resistance to TEC family kinase inhibitors.
