transgenic cotton strains, cry1f and cry1ac, and their event-specific identification. The present invention relates to plant breeding and the protection of plants against insects. More specifically, this invention includes unusual cotton plant transformation events comprising one or more polynucleotide sequences, as described herein, inserted within a specific site or sites within the genome of a cotton cell. in highly preferred embodiments, said polynucleotide sequences encode the "combined" lepidopteran, cry1f and cry1ac insect inhibitory proteins. however, the subject invention includes plants that have a single cry1f or cry1ac event as described herein. Additionally, the invention relates to cotton plants derived from this transformation event and assays to detect the presence of the event in a sample. More specifically, the present invention provides DNA assays and related assays for detecting the presence of certain insect resistance events in cotton. The assays are based on the DNA sequences of recombinant constructs inserted into the algadão genome and the genomic sequences that flank the insertion sites. These sequences are unique. Based on these insertion and boundary sequences, specific event initiators were generated. pcr analysis demonstrated that these cotton strains can be identified in different cotton genotypes by analyzing the pcr amplicons generated with these event-specific primer sets. therefore, these procedures and other related procedures may be used to uniquely identify these cotton strains. Kits and conditions useful for conducting the tests are also provided. These materials and methods can also be used to aid grafting programs in order to develop traces on cotton.
