The present invention relates to a mutant of 2-deoxyribose 5-phosphate aldolase and, more specifically, to a mutant of 2-deoxyribose 5-phosphate aldolase derived from a Haemophilus influenza Rd KW20 strain, to a gene for coding the 2-deoxyribose 5-phosphate aldolase mutant, or to a recombinant microorganism induced with the gene and a manufacturing method of a 2-deoxyribose 5-phosphate aldolase mutant for culturing the recombinant microorganism. According to the present invention, the 2-deoxyribose 5-phosphate aldolase mutant is useful for the manufacture of lactol or the like, comprising antiviral nucleoside, an anticancer drug, and a statin-based anti-hyperlipidemic drug, with excellent activation and low substrate inhibition with respect to acetaldehyde.
