The present invention relates to a glucose detection plasmonic biosensor having a groove slit groove structure. The biosensor includes a metal thin film which is formed on a substrate made of a transparent material. The metal thin film is provided with a slit providing a light path allowing light input from the upper portion of the metal thin film to be input to the substrate and n grooves on one side with reference to the slit at the upper portion of the metal thin film, and is provided with m grooves on the other side (n and m are natural numbers and satisfy n+m = 3). The biosensor includes a multi-slit groove structure also provided with one or more slits. The biosensor includes a grid slit groove structure forming a vertical groove slit groove structure and a transverse groove slit groove structure by crossing the groove slit groove structures.
